## What is Tafamidis?

Tafamidis (brand name: Vyndaqel) is a transthyretin stabilizer. It is prescribed to treat treat transthyretin amyloid cardiomyopathy (attr-cm; a condition in which a protein (transthyretin) builds up in the wall of the heart making it thicker which makes it difficult for the heart to pump blood adequately). Across the United States, 1,000 doctors prescribe Tafamidis to approximately 3,000 Medicare patients.

Tafamidis is primarily prescribed for the treatment of treat transthyretin amyloid cardiomyopathy (attr-cm; a condition in which a protein (transthyretin) builds up in the wall of the heart making it thicker which makes it difficult for the heart to pump blood adequately).

## Mechanism of action

Tafamidis is a selective stabilizer of TTR. Tafamidis binds to TTR at the thyroxine binding sites, stabilizing the tetramer and slowing dissociation into monomers, the rate-limiting step in the amyloidogenic process.
